# Creating new layers

You start most of your image editing in Photoshop by creating new layers. If you select the Layers palette from the top of your workspace and start to add new layers, you’ll see a dialog box asking you to name the layer.

After you click OK, you have a new layer on the screen, as shown in Figure 2-3. When you create a new layer, it appears on your working area.

To investigate the time course of changes in the number of peritumoral infiltrate of immune cells (IC) in metastatic melanoma patients treated with nivolumab alone. A total of 33 patients with advanced melanoma received nivolumab monotherapy in the first-line setting. The number of IC per HPF was assessed by immunohistochemistry in tumor samples obtained at baseline and 4, 8, 16 and 24 weeks from the start of the therapy. Overall, we observed a progressive fall in the number of IC during the course of treatment. IC per HPF decreased at 4 and 8 weeks from the initiation of nivolumab in accordance with the initial immune-related adverse events (irAEs) and tumor response. This change in the number of IC was correlated with significant differences in the evolution of irAEs in patients who experienced a clinical benefit (CR + PR, n=11) compared to those who experienced stable disease or progressive disease (SD/PD, n=8). A progressive and significant decrease in the number of IC per HPF was observed in melanoma patients who received nivolumab alone and it correlated with irAEs and tumor response.
